Job Description Summary:
The Scientist I, Multiomics Analysis will use cutting edge imaging and analytical techniques to map the in situ localization of translational biomarkers and therapeutic agents in preclinical animal tissues and clinical biopsies. The candidate will join a diverse team responsible for generating data to help elucidate mechanism of action, confirm target engagement, discover new targets, and probe underlying disease pathology. This position will join a diverse team employing Multiomics approaches to advance innovative therapies in neurodegenerative, ferroptosis, mitochondrial dysfunction, and inflammation.
The Scientist I, Multiomics Analysis is responsible for planning and performing scientific experiments that contribute to PTC’s research and drug discovery activities. The incumbent works cross-functionally with internal departments, and external resources as appropriate, as part of PTC’s discovery science project teams.
This position will adhere to relevant regulatory requirements and company Standard Operating Procedures (SOPs) as appropriate.

ESSENTIAL FUNCTIONS
Primary duties/responsibilities:
Demonstrates subject matter expertise and implements detailed instrumental workflows for internal imaging modalities.
Performs multi-modal imaging experiments (i.e. imaging mass spectrometry, IF, IHC) to visualize both biomarker and drug distribution in tissue sections.
Responsible for cryo-sectioning and thaw-mounting of fresh frozen tissues.
Perform rapid H&E and/or Cresyl Violet staining to quickly assess tissue coordinates and architecture.
Develops novel imaging methods (i.e. on-tissue derivatization) for the selective enhancement of analytes with poor sensitivity or low abundance.
Performs co-registration, ROI selection, and statistical analysis of digital images. Utilizes segmentation and pathway analyses to correlate biological changes with disease phenotype and/or drug administration.
Represents the organization in external collaborations and at scientific conferences. May participate in the authorship and publication of scientific papers and patents.
Interviews and contributes to the evaluation of candidates. Acts as a resource and trains junior staff. May direct and supervise the activities of other lab personnel.
Abides by all safety and regulatory guidelines. Maintains a detailed laboratory notebook.
Communicates effectively with peers and presents data at weekly group meetings.
Demonstrates compliance with corporate, department, and job-specific requirements.
Performs other tasks and assignments as needed and specified by management.
KNOWLEDGE/SKILLS/ABILITIES REQUIRED
Extensive hands-on experience using MALDI mass spectrometers and/or other ion sources (e.g. MIBI, DESI, SIMS, etc.) amenable to imaging mass spectrometry.
Experience with IF/IHC tissue staining protocols.
Expertise with Fiji, QuPath, Zen, and/or SCiLS image analysis software packages.
Proficient at cryo-sectioning.
* Minimum level of education and years of relevant work experience.
* Special knowledge or skills needed and/or licenses or certificates required.
Experience with brightfield and fluorescence microscopes or slide scanners (preferably from Zeiss).
Prior use of HTX instrumentation for MALDI matrix application.
Detailed knowledge of tissue staining protocols for IHC/IF.
Excellent verbal and written communication.
Detail-oriented worker with superb organizational and time management skills.
Analytical thinker with excellent problem-solving skills.
*Special knowledge or skills and/or licenses or certificates preferred.
Experience with laser capture microdissection is a plus.
Prior use of spatial transcriptomic instrumentation (e.g. 10X, Nanostring, etc.) is a plus.

*Expected Base Salary Range: $100,000 - $128,000.The base salary offered will be contingent on assessment of candidate education level, background, and experience relative to the requirements of the position they are being considered for, as well as review of internal equity. In addition to base salary, PTC employees are also eligible for short- and long-term incentives. All eligible employees may also enroll in PTC’s medical, dental, vision, and retirement savings plans.
